| Description | 3-Deazaneplanocin A, also known as DZNep and NSC 617989, is an inhibitor of the lysine methyltransferase EZH2 (KMT6). It is a cyclopentenyl analog of 3-deazaadenosine, originally synthesized as an inhibitor of S-adenosyl-L-homocysteine hydrolase. The inhibitor has been shown to decrease EZH2 levels and to inhibit trimethylation of lysine 27 on histone H3 in cultured human AML HL-60 and OCI-AML3 cells. It has the same effect on primary AML cells in a dose-dependent manner. 3-Deazaneplanocin A can increase expression of cell-cycle regulators p21, p27, and FBX032, which can lead to cell cycle arrest and apoptosis. | |
